Abstract:In this study, the preconcentration of Cr(III) ion in microorganism immobilized on perlite adsorbent by solid phase extraction was investigated. As the microorganism was tested Pichia kudriavzevii JD2. Cr(III) determination was made by flame atomic absorption spectrometry. The effects of pH, amount of adsorbent, volume and type of recovery solution, and flow rate and volume of the sample solution on the recovery efficiency of Cr(III) ion were investigated. By applying the column technique, the optimum preconcentration conditions were determined as pH 4, recovery solution 2 mol/L HNO3 and volume 10 mL, and sample flow rate 1-3 mL/min. Under the optimum conditions, the recovery efficiency of Cr(III) ion was found to be 100,1±0.3% of perlite immobilized on Pichia kudriavzevii JD2 with a 95% confidence level. Analytical variables with limit of detection (LOD) of 4.8 µg/L and limit of quantification (LOQ) of 15.8 µg/L were determined for the Cr(III) ion. The accuracy of the method was determined using standard reference materials (CRM). The relative error of the recovery efficiency was determined to be less than 10%. The method was applied to the determination of Cr(III) in various water samples such as tap water and mineral waters.
