Abstract:

The aim of the thesis study is to develop and characterize the PCL / PEO / HNT composite nanofibers with/without doxycycline loaded by electrospinning method and to determine drug release profiles. In the study, PEO at rates of 0.5, 1.0 and 1.5% (w / w) were added to the PCL solution at a concentration of 18% (w / v), respectively. Three different percentages of pure HNT as 0.1%, 0.3 and 0.5 (w / w) respectively, were added to the mixture of 18% PCL / 1.0% PEO, which was the best mixture in terms of morphological considerations. Then, two different surface modified HNT (HNT-APTES and HNT-GPTMS) were added to the composite mixture (18% PCL / 1.0% PEO / 0.3% HNT) with the same ratio of pure HNT. Moreover, each selected mixture which is resulted with smooth morphology was synthesized by addition of 5% DOXH and the drug release profiles were compared. For each of the mixtures prepared to ensure uniform fiber formation, the applied voltage, the distance between the collector plate and the needle and the flow rate, which are the parameters of the electrospinning process, were optimized. The physical and chemical properties of the drug loaded and unloaded nanofiber mats were investigated by Scanning Electron Microscopy (SEM), Fourier Transform Infrared (FT-IR), and Atomic Force Microscopy (AFM) methods. In consequence of the experimental studies, fiber diameters enhances from nanometers to micrometers as a result of modified and unmodified HNT addition into polymer solution. APTES-modified HNT additon was resulted in a high rate of burst release. On the other hand the GPTMS-modified HNT additive shows a controlled release profile, although the rate of burst release is elevated in comparison of unmodified HNT addition.
