Abstract:

In this study, we aimed to investigate the protective effects of hydrogen-rich saline solution (HZSS) on penile tissue in rats who underwent an experimental Mathieu type penile flap with penile tourniquet. In this study, 36 male Winstar-albino rats were randomly selected and divided into six groups. No operations were performed on the sham (S) group. In the other five groups, penile tourniquet (Pt) was performed with Mathieu type penile flap. PT10 and PT30 groups were performed Pt for 10 and 30 minutes, respectively. In the HZSS10 and HZSS30 groups, intraperitoneal HZSS 5ml/kg was given before opening the 10 and 30 minutes of Pt. All rats were sacrificed after 4 hours and penile tissue samples were taken. Biochemically, GSH, MPO, MDA, IL-6 and TNF-α values were measured. Tissue damage and apoptotic index were examined histopathologically. Results were evaluated by one-way ANOVA and Pearson's chi-square tests. Biochemically, MDA, MPO and IL-6 values were significantly decreased in favor of HZSS groups (p <0.05), while GSH values were increased in HZSS groups in contrast to PT groups (p> 0.05). While TNF-α values were lower in HZSS groups, there was only significant difference between PT¹⁰ group and HRSS¹⁰. Inflammation was significantly decreased in treated groups compared to the PT groups (p <0.05). Apoptosis was not detected in any group. As a result, hydrogen rich saline solution showed protective effect on penile tourniquet application after hypospadias model prepared by penile flap, reducing oxidative stress, inflammation and decreasing inflammation histopathologically
