Abstract:In our study, there are 50 patients who are recently diagnosed as having Type 2 Diabetes and 30 healthy individuals. Patients were divided two groups and three –months cure has been carried out with Rosiglitazone and Metformin. Before and after the cure, levels of catalase, Superoxide dismutase (SOD), total antioxidant capacity (TAC), malondialdehyde (MDA), 8-hydroxy -2'- deoxyguanosine (8-OHdG), paraoxonase and leptin are measured. Besides, effects of factors such as age, history of family, smoking status, physical exercise on oxidative stress parameters and leptin were statistically evalvated. Before the treatment while SOD activites and TAC levels of the total patient group are found to be considerably low compare to the control group (respectively, p=0.005, p=0.010). MDA levels are significantly high (p=0.024). Values for Catalase, 8-OHdG, and paraoxonase of the total patient group hasn‘t demonstrated any difference compare to the control group. While TAC levels of Rosiglitazone group before the treatment were significantly lower than the control group (p=0.015), but MDA levels were higher than the control group (p= 0.019). After three months of treatment, TAC levels are found to be considerably lower than (p=0.027) the control group. MDA and 8-OHdG levels before the treatment have dramatically decreased after the treatment (respectively, p=0.008, p=0.000). SOD activities of Metformin group before treatment were considerably lower than (p=0.000) the control group. While TAC levels after treatment were significantly higher than the control group (p=0.000), SOD activities were considerably lower than the control group (p=0.001). After the treament, increase of TAC levels and SOD activities has been observed when compared to before the treatment valves (respectively, p=0.000, p=0.029). While decreased blood sugar and HbA1c has been observed in both treatment groups after the treatment, in Metformin group significant decreases has been observed on total cholesterol, LDL – cholesterol and triglyceride levels (respectively, p=0.017, p=0.004, p=0.200). Significant correlations have been found between pre-treatment catalasa and 8-OHdG values (p=0.003) and post treatment TAC and SOD values (p=0.020) in Rosiglitazone group. Significant correlations have been found between pre-treatment TAC and SOD values (p=0.005) and post treatment catalasa and TAC values (p=0.006). In control group, significant correlations are observed between catalasa and 8-OHdG values, catalasa and MDA values (respectively, p=0.013, p=0.016).
