Prognostic importance of arid1a protein loss ġn colorectal cancer

Abstract:

Colorectal carcinoma is one of the most common malignancies in our country and worldwide. It is still a cause of high mortality and morbidity for both genders. T-N-M staging is the gold standard prognostic parameter for colorectal carcinoma. However, even in the same stage, the prognoses of patients vary enormously due to molecular differences. One hundred and forty-two colorectal carcinoma cases, diagnosed at the Pathology Department of Gazi University Medical School, were included in this study between the years 2008 and 2011. We applied the ARID1A immunohistochemical marker to paraffin tissue blocks of 142 patients. Our aim was to analyse the immunohistochemical expression of ARID1A in resection materials and evaluate its correlation with patients' clinical features (age, gender), tumour localization, tumour dimension, macroscopic growth pattern, histologic type, grade, angiolymphatic invasion, T-N-M stage, liver and other distinct metastasis at the time of diagnosis or during follow-up, local recurrence and overall survival. This way we planned to determine the prognostic significance of this immunohistochemical marker. Fourteen point seven percent of the cases showed loss of nuclear ARID1A expression. ARID1A loss was more common in early stage tumours than late stage tumours. Immunohistochemical expression of ARID1A were not related to the other major prognostic parameters including T-N-M categories, angiolymphatic invasion and overall survival. This study showed that, prognostic significance of ARID1A is limited and it's not a poor prognostic marker in colorectal carcinoma. In conclusion, to clarify the conflicting results in the literature, and to determine the role of this marker for prognosis and treatment, there must be larger studies with standardized technical methods.
