Abstract:

The aim of this thesis is to develop, characterize and evaluate the in vitro efficacy of PLA/PVP composite fibrous scaffolds containing (-OH) or (-COOH) functionalized CNT for bone tissue engineering. Firstly, the solution containing 10% (w/v) PLA and 2% (w/v) PVP polymers was prepared in a solvent mixture of tetrahydrofuran (THF): dimethylformamide (DMF) at a ratio of 70:30 (v/v) as a control group. Then, to this polymer mixture, CNT-COOH or CNT-OH was added in the ratio of 0.25%, 0.5% and 1.0% (w/w of the total polymer mass) to produce composite fibrous scaffolds. Morphological, physicochemical and mechanical properties of fibrous scaffolds were determined by scanning electron microscopy (SEM), atomic force microscopy (AFM), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), surface contact angle, hydrolytic degradation, and tensile strength analyses. According to SEM and AFM results, all scaffolds were produced with bead-free and uniform morphology. FTIR spectra show that all scaffold components retain their characteristics; XRD diffraction patterns showed that PLA showed prominent features and all scaffolds were amorphous. The hydrophilicity and tensile strengths of scaffolds containing both CNT-COOH and CNT-OH increased in direct proportion to their concentration. As a result of hydrolytic degradation, the highest mass loss was observed at 74.4% at the end of 63 days on the scaffold containing 1.0% CNT-COOH. MTT analysis of 1, 4 and 7 days showed that all scaffolds had no toxic effect on MC3T3-E1 cells and scaffolds containing CNT-COOH allowed higher cell proliferation than scaffolds containing pure PLA/PVP and CNT-OH. In conclusion, CNT-COOH is the most suitable candidate for tissue engineering applications.
