A Simple Method to Prepare an Organ Preservation Solution

Özet

Objective: The purpose of this study was to provide an inexpensive and easy method to prepare organ preservation solution. Material and Methods: The organ preservation solution was prepared by using solutions and drugs, which are still used in clinical practice. Twenty-four rat heart-lung blocks were removed and were randomly divided into 4 groups (n = 6). In the control group, lung blocks were perfused with isotonic saline solution and were immediately processed for examinations. In the other three groups, lung blocks were perfused with low potassium dextran (Perfadex), University of Wisconsin (UWS) and the solution prepared by us (GOPOKS), respectively and they were preserved in these solutions for 24 hours. Cell death, degeneration, cytokin level, lipid peroxidation and antioxidant enzyme activities were studied, and the costs were evaluated. Results: No difference between the groups was found regarding severity of cell death and degeneration (p > 0.05). IL-8 level was significantly higher in the UWS group than in others (p < 0.05); however, there was no difference between Perfadex and GOPOKS. SOD activity was significantly lower in the GOPOKS as compared to others (p < 0.05). In addition, the cost of GOPOKS was much lower than that of Perfadex and UWS (25-30 TL/L vs. 300-400 L/lt and 700-800 TL/L, respectively). Conclusion: In this study, as an organ preservation solution, GOPOKS proved as good as Perfadex and UWS. GOPOKS provided better preservation against oxidative stress as compared to the others. Additionally, GOPOKS was much cheaper than other organ preservation solutions examined in this study. According to initial results, GOPOKS can be used in experimental studies without any concern; however, further studies are required before use in clinical practice.
