Özet

In this study, adenosine deaminase enzyme which plays an important role in cancer metabolism was isolated and partly purificated from cattle liver, and its kinetic properties were established with and without turmeric extract. V max and Km values of ADA enzyme were found to be 0.80 absorbance unit (as specific activity 40 mIU/g protein) and 100 mM respectively by using Michaelis Menten and Lineweaver Burk graphs drawn in the studies without inhibitor. In the Michaelis Menten and Lineweaver Burk graphs drawn with two different concentrations of Curcuma Longa extract, Km value was found to be 100 mM at both concentrations. V max value was found to be 0.5 (25 mIU/g protein) at the 10 % turmeric concentration, but it was 0.3 (15 mIU/g protein) at 20 % turmeric concentration. These results show that turmeric constituents inhibit ADA enzyme non competetively. This kind of inhibition of ADA enzyme by turmeric extract might be one of the mechanisms contributed to anti cancer potency of curcuma longa.
